Phosphate-Buffered Saline with Tween 20 (10x stock, 1L)
Alias: PBST
Reagent	Amount	Final Concentration
NaCl	80 g	1.37 M
KCl	2.0 g	27 mM
Na2HPO4	14.4 g	100 mM
KH2PO4	2.4 g	18 mM
Tween-20	20 mL	2%
Directions Mix all of the above with 800mL ultra-pure water and dissolve well till no particle observed. Adjust pH to 7.2, 7.4 or 7.6 with 5M HCl. Then add ultra-pure water till 1L.
Sterilization PBS can be sterilized by autoclaving for 20 min at 121°C or 15 psi (1.05 kg/cm2). PBST may be filter with 0.22um membrane filter. TWEEN-20 is not stable to autoclaving, particularly with metal cations in buffer solutions. TWEEN-20 is heat sensitive and will darken when exposed to elevated temperatures.
